The researchers propose to utilize the next-generation sequencing platform, 454 Roche DNA sequencing technology to establish a method to analyze mixed, limited, and/or degraded DNA samples using mtDNA and miniplex STR markers simultaneously. This project will result in two deliverables: (1) a validated "front-end" system targeting both nuclear STR and mtDNA markers for use with the 454 sequencing technology for application to forensically relevant samples (i.e. degraded and mixed); and (2) Software for the interpretation of data generated by the system.
