Network Biosystems (NetBio) previously developed (based on NIJ-funded work performed by the Whitehead Institute) Genebench-FX Series 100, a microfluidic-based instrument for separation and detection of STRs, mini-STRs, and mitochondrial DNA sequences. The goal of this project is to develop a module for extraction and purification of DNA from blood, bloodstains, and buccal swabs in under fifteen minutes in a disposable plastic biochip. The research program involves the analysis and characterization of several methods of DNA extraction and purification to assess their performance and compatibility with NetBio's plastic biochip and microfluidics technologies. The most promising approaches will be optimized and modifications will be made to the chips, coatings, and reagents so that the extraction/purification module attains the required performance levels. Crime laboratory practitioners will provide technical guidance and feedback, as well as appropriate samples, including non-probative casework samples, for testing of the DNA extraction and purification methodologies developed in this program.
